Studies on the effect of Diuretics to the Biopharmackokinetics of Sulfadiazine


Abstract
For the purpose of investigation in the process of absorption, distribution, metabolism, and excretion, sulfadiazine was administratrated with Diuretics such as Hydrochlorothiazide, Furosemide, and Acetazolamide.
These studies were made on the biopharmacokinetics of Sulfadiazine by examining the serum concentration of rabbit which had been administrated them alone or compounded with Diuretics.
The results were summarized as follows:
1. When Sulfadiazine concomitant administrated with Diuretics, the serum concentration of Sulfadiazine was increased by Furosemide or decreased by Acetazolamide. Therefore, elimination rate of Sulfadiazine was reduced by Furosemide and accelerated by Acetazolamide, Hydrochlorothiazide. The Biological half-life of Sulradiazine prolonged by Furosemide and reduced by Acetazolamide, Hydrochlorothiazide, ADC of Sulfadiazine was increased by Furosemide, Hydrochlorothiazide and decreased by Acetazolamide.
2. When the Sulfadiazine administrated 2 hours before or after with Diuretics, elimination rate, half-life and AUC of Sulfadiazine was no difference.
3. When the Acetylsulfadiazine, which is Sulfadiazine metabolite, was measured, metabolism rate of Sulfadiazine was increased by Furosemid, Hydrochlorothiazide and decreased by Acetazolamide.
